Spectral Flow Cytometry with Cytek Aurora
Spectral flow cytometry captures the entire emission spectrum of fluorochrome dyes, which permits the identification of multiple individual dyes in a single sample even if they have overlapping spectra. This advanced technology pemits multi-color staining of individual samples with excellent resolution. Because detectors are highly sensitive, antibodies can be used in lower concentraion than needed for standard flow cytometry. In addition, autofluorescence can be substracted. CyTOF
Mass cytometry (CyTOF) is a variation of flow cytometry in which antibodies are labeled with heavy metal ion tags rather than fluorochromes. CyTOF is capable of generating data on 48 parameters for each event passing through the detector. The workflow and operation are very similar to flow cytometry, but rather than using fluorescence, elemental tags are utilized. This can be very useful if you have a limited amount of input sample and want to extract as much information as possible in a single experiment. The Liver Immunology Services group within the LCIAI has experience applying CyTOF to studies of liver diseases.
There are currently two Fluidigm CyTOF2 instruments within the Parnassus Flow Cytometry Core. They are operated using CyTOF Acquisition Software and are available for all labs to use, following appropriate training.
